Fingerprints of Polysaccharides by Recombinant Lectin Receptor-Fc Fusion Proteins: Towards the Treatment of Infectious Diseases and Autoimmune Diseases
Full description
Introduction/Background
Lectin contains the carbohydrate recognition domain (CRD) and has the ability to bind specific sugar epitopes on polysaccharides or glycoprotein. The effective components of many herb drugs rely on polysaccarides, which activate immune functions via binding to lectin and other receptors.
Aims/Hypothesis
Currently, the characterisation of polysaccharides is based on enzyme digestion and mass spectrometry analysis. The current method is complicated and very expensive. In addition, the potential polysaccharides functions analysed by the current method are not informative.
Research
We have developed a high throughput technology to apply the recombinant lectin fusion proteins to analyse the components of polysaccharides and glycoproteins. This technology can be used for the quantitative and qualitative analysis of polysaccharides and glycoproteins contained in the samples, for the screening of active substances contained in the samples or within the polysaccharides and glycoproteins for activation or suppression of immune functions, for the screening of receptors for virus, bacteria, fungus, and other polysaccharides-containing or glycoprotein-containing organisms, for the purification and removal of polysaccharides and glycoproteins capable of binding with these receptors directly or indirectly, to use the recombinant proteins for the treatment of infectious diseases and autoimmune diseases and to generate mAbs against these lectin receptors for the treatment of infectious diseases and autoimmune diseases.
Conclusion
We have developed a high throughput technology to apply the recombinant lectin fusion proteins to analyse the components of polysaccharides and glycoproteins.
